On September 25, 2010, Killam joined the cast of the NBC sketch comedy/variety show, Saturday Night Live, for its 36th season. He was a prominent member of the SNL cast for six years before he departed in August 2016 to pursue other interests.

Killam went on to make numerous guest appearances in TV shows like Community, iCarly, New Girl, Robot Chicken, and A.P. Bio.

He also had movie appearances in such diverse movies as Grown Ups 2, Underdogs, 12 Years a Slave, Teenage Mutant Ninja Turtles, and Ted 2.

He also had recurring appearances as Gary Blauman in How I Met Your Mother, sharing the screen with his wife, Cobie Smulders.
