Ross grew up alongside his younger sister Robyn in his hometown. He went to Jonathan Dayton High School. He then enrolled at Boston University, where he majored in broadcasting film and minored in political science. He graduated from the Boston University College of Communication in 1987.

In 1994, Jeff began performing stand-up at open mic nights at The Boston Comedy Club in New York City. By 1995, Ross performed at comedy clubs like the Comedy Cellar and Stand Up NY. The same year, he was invited to participate in a roast of actor Steven Seagal by the New York Friars Club.

He became a regular at the Friars Club roasts and was given the title of Roastmaster General. In 1999, comedian Jimmy Kimmel hired him as a writer on The Man Show, and he wrote some of the punchlines for Billy Crystal’s monologue at the Oscars in 2000.
He became a roaster at all Comedy Central roasts in the 2005 roast of Pamela Anderson. He also roasted Charlie Sheen dressed as Muammar Gaddafi; Rob Lowe dressed as Purple Rain-era Prince, roasted Roseanne Barr dressed as the late football coach Joe Paterno.
Some of the other renowned celebrities Jeff roasted are William Shatner, Larry the Cable Guy, Joan Rivers, Donald Trump, James Franco, The Border, Bruce Willis, and Alec Baldwin.
In 2005, Jeff released the film Patriot Act: A Jeffrey Ross Home Movie, which he both directed and produced and which documented his experiences on the Iraq tour.
In 2009, he published his first book entitled I Only Roast the Ones I Love Busting Balls Without Burning Bridges. His book is about his journey to becoming Roastmaster General and includes tips on writing roast jokes and how to put on a roast.

In 2018, Jeff and fellow comedian Dave Attell embarked on their Bumping MMics comedy tour. Footages of their tour were released in a three-part special on Netflix. The Bumping Mics comedy special filmed during three of their performances at the Comedy Cellar and consist of observational and political humor interspersed with Ross and Dave roasting one another as well as members of the audience.
As of 2019, Netflix began airing the series Historical Roasts where Jeff hosts fictionalized versions of comedy roasts for figures like Anne Frank and Abraham Lincoln.
